Release a cat or a fox into the market and see who chases it.

Set up a stall in the market a sell scooby snacks. Werewolves can't resist.

Or lastly, kill them all and let Erastal sort them out

You know how dogs in the real world can sniff out diseases/drugs/etc? Train a dog to sniff out lycanthropes.

Silver Crusade

Test for that wet dog smell...
or walk around with a female wolf in heat, that should bring out the male werewolves.

While it is often considered a faux pas to simply ask if someone is a werewolf, "Never Have I Ever" is a commonly accepted game.

Start off with a few possibly scandalous statements, and lead up into "Never have I ever turned into a turned into a bloodthirsty wolf when my twice-damned flesh was exposed to the light of the full moon."

If no one puts down a finger, roll Sense Motive. Cheating is simply deplorable.

Edit: Just noticed this thread was necro'd from 2011. Also deplorable, let sleeping dogs lie.

Less common signs of a lycanthrope from folklore that I can remember, many not quite sensible:

Unibrows.

Hairy palms, also falsely purported to be a symptom of something else.

Having the middle and index fingers be the same length.
