Will any retroactive changes be coming down the line with the rules for large PCs? Like would a Gnoll with Giant Gnoll ancestry actually be large now? What happens with a Large sized Barbarian PC who has the Giant Instinct? Are they now using a even bigger weapon than large? If not, wouldn't they actually have no penalties for using a large weapon since it is in fact, normal for them?

While I doubt that we'll see changes like that to hit Legacy content, you do now at least have something to point to if you and your GM would like to houserule something for older heritages.
As for the barbarian, that's already called out in the original Core Rulebook:
You can use a weapon built for a Large creature if you are Small or Medium (both normally and when raging). If you're not Small or Medium, you can use a weapon built for a creature one size larger than you. You gain access to this larger weapon, which can be of any weapon type otherwise available at character creation. It has the normal Price and Bulk for a weapon of its size. When wielding such a weapon in combat, increase your additional damage from Rage from 2 to 6, but you have the clumsy 1 condition because of the weapon's unwieldy size. You can't remove this clumsy condition or ignore its penalties by any means while wielding the weapon.

EDIT: I will say that there could be some improvement room for things like Giant's Stature and Titan's Stature. if only because this would create an actual feat tax for Large-sized giant instinct barbarians. A personal preference would be for Titan Stature just to get a "or you are a member of a Large-sized ancestry" added to the requirements while allowing them Giant's Stature's "action". That seems to just tie everything up without making everything too messy.

I believe changelings stayed uncommon, which suggests keftiu was off base with that specific statement. Also minotaurs and centaurs are only uncommon, not rare.
I still doubt gnolls are changing. Letting other uncommon ancestries become large with specific heritages just seems like a GM approval headache. Besides, I've never heard of a large sized gnoll outside of some specific demonic enhancements on NPCs.
