I find it unlikely that they would base a class around a single weapon. The concept of truth may be at its core, though. I think it may be a psionic leader class. The monk is a psionic striker and the psion is a psionic controller. If we get a psychic warrior or soulblade, it may be a defender. The seeker class being a divine one also has some merit. The seeker could be a divine leader that uses divine magic to find the way. We will have to wait and see.

I cannot tell if you got the joke or not, but it also has alot of merit being an arcane class that flies around seeking things as well. ;)

While it may be a new Psionic class, remember that the PH3 is adding Psionic, Divine, and Primal classes to the game.
Seeker sounded Divine to me, although you could construe the name to be anything I suppose ...
the Harry Potter Reference and the Sword of Truth book references are now lost. :(

Yeah, the name could apply to enough things that I can see any number of options. Here are my theories, in order from most likely to least likely.
Psionic Seeker: Gifted at clairvoyance, premonition and awareness of the world around them, a psionic warrior that is able to predict their enemy's moves and always be one step ahead of danger - serving both as a psionic defender, and filling the role for the 'clairsentience' discipline that Psions no longer have.
Primal Seeker: A primal hunter that chooses their prey and brings them down no matter what, which would fit well as a striker - possibly a ranged striker, which primal currently lacks.
Divine Seeker: A seeker of knowledge and wisdom, or possibly a form of inquisitor that hunts those that are enemies of the church - again, a ranged striker would seem appropriate, and is currently absent for divine.
My bets on the Psion, but I can certainly see it falling into any of these categories... or being something else entirely. I didn't really see the Invoker or Avenger coming with PHB2, yet now that they are around, they seem a perfectly natural part of the game.

That's one reason why I am suspecting the Psion Defender - the current balance of roles is slightly Striker heavy, so if the PHB3 sticks to 1 Striker, we will be close to a near-perfect balance between the roles. And we already know the Monk is a Striker, so if there was just 1, that would be it.
But... we'll see whether that holds true. Strikers tend to be popular and easy to fill up on compared to other roles, and possibly easier to have multiples of within a power source. Could they produce another Divine Leader while keeping it distinct from the Cleric?
Part of the problem is how several of the initial classes were a bit too expansive - Clerics being able to go melee or ranged, for example. If you had kept the default cleric as the Wisdom-based ranged character, you would later have room for a Divine Leader called a 'Templar' or some such - which would exist as the Strength-based battle cleric role.

With the name Seeker I doubt this will be the case, but I would like to see another Controller. I mean, right now we have only four controllers in the whole bunch, IIRC. In fact I can only think of three off the top of my head, druid, invoker, and wizard. Another controller would be nice.
You are right that there are three. Psion will be a fourth (already announced), but perhaps the PHB3 will have 2 controllers, instead of just one.

One possibility that only just came to mind - if the Seeker ends up being a Divine class, and is some form of inquisitor or similar concept... I can see that being a very viable way to create a melee Controller.
We currently have the druid which is a blend of melee and ranged controller ability. I can see a new class this is entirely up close, inflicting status effects and conditions on nearby foes, and possibly having various ways for punishing enemies that attack them directly (to ensure their survival in melee.)
Just a thought, but given they are trying some new things here, I can see this as a possibility.
