There's a lot of Fortitude saves going on in this latest post.
EVERYONE has to make a Fortitude DC 16 vs. sickened 1 (2 on a CF) first due to the xulgath stench.
Rulean, Clover, and Snowy then need to make a Fortitude DC 16 vs. Slow 1 (2 on CF) and also take -2 to Fortitude saves vs. poison for 1 minute.
Clover then needs to make two Fortitude DC 18 saves vs. poison.
Failing 1 makes her Fatigued.
Crit Failing 1 makes her Fatigued and Unconscious.
Failing 2 makes her Fatigued and Unconscious.

Correct. The secret door is inside the arc of the runes on the floor.
The door is not on Harlock's map, but you could come to the conclusion that the door connects the two chambers with runes on them somehow.

I mass rolled the stench to keep things moving. Everyone who might want to use a Hero Point to avoid the Sickened 1 condition has one to use, if you choose. Feel free to roll that at the top of your post, if you want to use it.

Enfeebled 1
You're physically weakened. Enfeebled always includes a value. When you are enfeebled, you take a status penalty equal to the condition value to Strength-based rolls and DCs, including Strength-based melee attack rolls, Strength-based damage rolls, and Athletics checks.
Frightened 2
You’re gripped by fear and struggle to control your nerves. The frightened condition always includes a value. You take a status penalty equal to this value to all your checks and DCs. Unless specified otherwise, at the end of each of your turns, the value of your frightened condition decreases by 1.
Sickened 1
You feel ill. Sickened always includes a value. You take a status penalty equal to this value on all your checks and DCs. You can't willingly ingest anything—including elixirs and potions—while sickened.
You can spend a single action retching in an attempt to recover, which lets you immediately attempt a Fortitude save against the DC of the effect that made you sickened. On a success, you reduce your sickened value by 1 (or by 2 on a critical success).
All three conditions impose Status penalties, so as I understand it I only take the greatest value of the penalties.
Sickened is all checks and DCs.
Frightened is the same.
Enfeebled is strength based attack, damage, and Athletics checks. It is more limited but is the only one that affects damage.
So next round, I will be at:
-2 to all checks and DCs
-1 to all Str-based damage
After that round, the frightened is reduced. On Round 3 it might be worth an action to get rid of the sickened condition. Assuming I don’t need to ingest anything, there is no benefit to doing it earlier than that.

Looks correct on your assessment of the penalties.
Something that I forgot: you take a -2 penalty to get rid of sickened while within 30 ft of any xulgath.
