Best is subjective here. I'm looking to make a discussion on the best 6 character combo for Wrath of the Righteous. Any character is up for inclusion, and as new class decks get released, feel free to add a new combo. So, the gist is this, given the challenges in WotR, what set of characters would be the best 6 character combination?
Personally, I've not figured out six yet, but I'd probably want to include the following:
WotR Kyra, for her power to use her Divine skill plus she can pack some Cure.
Adowyn, can scout and evade (and later let others evade too). Plus can pack a Cure.
WotR Seelah. Got to have a Paladin for Radiance, 1d6 to checks.
I'm still figuring out the other 3. I'd probably want to start diversifying skills with the remaining 3. So, what do you think?

Personally I think you have lots of options here. With that core group you've got healing, scouting, tanking covered. I'd want to cover Int/Arcane, so I'd toss in Enora, Balazar, or Seoni. Frankly, they're all great, even if I'd probably err in roughly that order. Balazar is probably the strongest / most reliable, even if I do like Enora more :)
I think Alain is great for a focused monster murderer, who at later levels can selfishly gain group synergy, so I'd include him.
For reference, my preferred group of 4 is Adowyn, Alain, Enora, and Kyra. For 5th I'd almost certainly pick Seelah. So I think our preferences are at least mildly aligned.
At that point, you can probably throw in anything you want. I think my vote last time was for Imrijka, but Shardra is very solid with a group of 6. Harsk can be very solid. You can double down on Arcane and have both Enora and Balazar, which is pretty boss. I _might_ replace whoever it was with Arueshallae not long after anyways. Or just use Ekkie.

You've got divine pretty well covered, with a mix of melee, ranged, and spell combat users.
The obvious gap is the Arcane skill. I'd propose Balazar, since he's from WotR, and is less susceptible to the Carrion Golem than other arcane casters. Sure, his Knowledge check isn't as good as Enora's, but it's at least respectable (and WotR Kyra's knowledge is surprisingly allright). Finally, a couple of things specifically reference summoners, so bonus points if he is around.

If it helps, here is my current 5-player group. We actually haven't failed a scenario yet, and we just finished Adventure B.
- Adowyn (to help with scouting and eventually helping others evade summoned banes.)
- Shardra (for scouting and also heavy support when dogpiling locations.)
- Balazar (because he's just a BEAST.)
- Crowe (to just smash the crap out of things.)
- Ekkie (to nullify locations that make you summon and encounter banes at the start of your turn.)
Some of them are changeable - I think you could swap Crowe for Alain or Seelah
For a sixth, I'd add Imrijka or Enora. Ekkie has been a champ in our game though, making the Guardpost a trivial location.

The big thing with a party of 6 seems to be the Armies. If I were making a 6 character dream team, I'd be pretty sure that for each army that exists, no character had to resort to d4s.
If I remember correctly, there is an army with both Ranged and Survival, so you'll want another character with Ranged or Survival. I haven't cracked open any of my Wrath decks yet, so someone who's more familar with the check breakdown can decide if a given party passes or fails the "no defaulting to a d4" test. And we won't know for sure until AD6 comes out that a given party will pass this test. :)

As far as the Adventure 2 armies go, though, what you're looking for to avoid defaulting to a d4 requires:
Diplomacy, Knowledge, Ranged, and Survival on different characters
Arcane, Divine, Knowledge, Perception, and Diplomacy on different characters
And Divine, Melee, Stealth, and Fortitude on different characters.
Starting with Hawkmoon's decent base of Kyra, Adowyn, Seelah, we see that the first army means we need another person with Ranged or Survival; the second requires an Arcane caster and someone with Perception; and the third means we're going to need another person with Melee or Fortitude.
Crowe actually looks like a great fit here - he has Arcane, Survival, and both Melee and Fortitude, so then you're almost free to fill in as you want. I'd probably take Harsk for Perception - it's better than Alain or Seoni's, plus he's better able to fight multiple times in a turn - and for the last one, I don't know, you have lots of great options. Shardra has the amazing-for-large-groups power to provide rerolls, so I'd probably end up going with her. Ekkie's tempting, though.

I'm playing a 6 character solitaire campaign and my party is as follows (all are WotR versions)
Alain. His ability to ride in and out of damaging locations is invaluable. Plus he can keep the abyssal rift temp closed by doing his horsey shenanigans. This is ignoring his ability to ride roughshod ( although Donohan is impeccably shod of course) over most any martial foe A rating
Imrijka. Her ability to chew through a monster heavy locale has been useful at times, as has her ranged weapons helping with hairy checks from afar. But overall meh. C rating
Balazar. A very capable character, and his knowledge stat and high charisma are both useful. B rating
Seelah Her Paladin's Helm has been an amazing boon in that it reflects all damage types on a reveal. Very tough and designed to take on the most numerous foes we face. B+ Rating
Kyra. Has been an invaluable support character so far. Her blessings largely go to aid others and then fuel healing by doing so. Not surprising if her turn starts with 0 cards in hand. Was invaluable in defeating the armies. A+ Rating
Seoni. Has trouble with more than a few of the banes, quite a capable spell aster but no synergies with the set ( One of her role cards changes all that ) C- Rating
