In honor of the website I'm frequenting, and a module I bought, I should give honorable mention to Imprisoned with the Pharoahs.
At least the OP didn't ask for very best of Lovecraft-inspired film, because there's an awful lot of pain out there...

My favorite adaptation would have to be 1970s "The Dunwitch Horror" starring Sandra Dee, Ed Begley (Sr.) as Dr. Armitage, and a very young Dean Stockwell as Wilbur. It may be a bit dated and B movie (or C movie), but I think you can tell they were really trying to create a real Lovecraft story (not just some schlock and bewbies).
Probably the best tribute to Lovecraft without being Lovecraft is "In the Mouth of Madness" by John Carpenter.

That's a pretty good one, easily the most professional.
While it's an appalling movie, I kind of love the basic idea behind From Beyond.

Also, feel free to choose a ghost-written work (like "Imprisoned with the Pharaohs").
The only catch to this thread is that the one work you chose must have been written by Lovecraft, not inspired by the Mythos--so, Tensor, while I love 'The Hounds of Tindalos', that story, and the collection surrounding it, was written by F.B. Long--so it doesn't count.
Maybe we should make a thread in the same vein, but choosing the one story inspired by the Mythos?

The Rats in the Walls
I'm terrible at picking favourites, but when I think Lovecraft, this is the first story that pops into my mind, and it wasn't even the first one I read (think that was Mountains of Madness) so it must have made the biggest impact
My brother recently began reading HPL, and read this one when his wife and son were out of town. When he goes to bed, he turns off the light and hears this weird scratching sound. Lights come on, the sound stops. This happened about two or three more times, by which point he is freaking out. Finally, he turns the lights on once more, and the noise doesn't stop - but this time there's a lump moving around under his covers!! He throws back the covers only to find - his mini Doberman (all of about 9 lbs), scratching and chewing on himself.

Best film? I'd have to say "Dagon" (watshisname Gordon's version of "The Shadow over Innsmouth"). The only other Lovecraft films I've seen, I've blocked out of my memory.
By the way, any fan of Lovecraft's should check out "Cthulhu 2000", an anthology of short stories written by fans of H.P. Lovecraft. The stories "The Barrens", "Fat Face", and "The Unthinkable".

Wow. Can't believe it took me this long to chime in!
My favorite Lovecraft story is probably "At the Mountains of Madness," I suppose. But there's about 20 other stories that are close seconds.
Favorite Lovecraft-themed story not by Lovecraft: "Notebook Found in a Deserted House," by Robert Bloch.
Best film? The silent movie version of "Call of Cthulhu" put out by the HPL Historical Society.

A wonderful tale, too. It will be available soon from Chaosium in the upcoming reprint of Bloch's collection Mysteries of the Worm. Until then, you can read it in Del Rey's Tales of the Cthulhu Mythos (a reprint of AH's 1990 book of the same title).
