DMPCing Krolmnite for the moment.
Krolmnite moves towards the vanguard and casts haste on Ava, Dashel, Duncan, Selveshanea, and Elesandira.
With startling speed, the ascomoid closes the distance between it and Duncan and attempts to run the dwarf over. Duncan, take 16 HP (but see below). Just before it impacts, it emits a foul-smelling jet of fine powder from one of the many craters pockmarking its surface; the jet spreads into a cloud around Duncan, Elesandira, and Selveshanea. DC 16 Fort saves* to avoid being nauseated while in the cloud.
Duncan, you have a choice, depending on whether you make your save. When it runs you over, you can elect to take the full impact, take an AoO, and try to hit it as it closes; you may also forgo the AoO and try to jump out of the way (i.e., take a DC 17 Reflex save for half damage). I think I know what you'll try to do ;-).

It's a trample, not a bull rush--essentially an autosuccess overrun that does damage:
As a full-round action, a creature with the trample ability can attempt to overrun any creature that is at least one size category smaller than itself. This works just like the overrun combat maneuver, but the trampling creature does not need to make a check, it merely has to move over opponents in its path. Targets of a trample take an amount of damage equal to the trampling creature's slam damage + 1-1/2 times its Str modifier.
Not that it matters.
Duncan swings at the giant rolling puffball as it runs him over, his pick finding some vulnerable spot in its peculiar anatomy. It collapses on him, oozing fetid fungal fluids on the dwarf.
Done with combat. Elesandira, make another Fort save; here's the description of the poison "Spores—inhaled; save Fort DC 16; frequency 1/round for 6 rounds; effect 1d2 Str damage; cure 2 saves.", and you've made one save already.

Duncan dismisses his bane effect as soon as he realizes he has won...and proceeds to extricate himself from the slop.
Freed, he moves a bit back towards his allies and starts casting CREATE WATER repeatedly over himself to wash the foul goop from his body and gear.
When she is close enough he asks, Eh, Eretria, ye ain't know that Prestidigiation spell do ye? I might be more pleasant company if ye do.
Regardless, once sufficient attempts are made to clean up, at least enough for him to not give his position away by smell, Duncan is ready to resume.

Ok, friend/mount/Duncan. Silf is used to funny smells from being locked in too tight cages with other familiar creatures wizard slaver kept. Did not clean often. Duncan was Very ferocious. Silf thinks Duncan makes a great warrior against big enemies but Duncan WAS hurt. Should Duncan wear better armor perhaps?}}

Dashel, even though you weren't familiar with the fungus the toxins it used were familiar enough that you're able to treat Elesandira so that she's able to resist them.
After curing Duncan up (I think he's the only one who took any HP) and cleaning the slime off of him (the perils of prestidigitation spells from Osiriani casters; now he smells like sandalwood and spices), the party continues towards Lightning's Call.
Time passes. In the lightless depths, it's not easy to tell how long it has been since encountering the ascomoid, but none of you are hungry or tired yet. Duncan and Elesandira, scouting ahead, spy at the edge of their vision another one of the giant puffballs, which begins to roll towards them.

Elesandira's first arrow goes wide of the onrushing puffball, but the second hits. The fungus does what the other did, releasing a cloud of toxic spores just before it hits Duncan. Elesandira is again sent away gagging, while the doughty dwarf disregards the cloud. The ascomoid runs over Duncan, inflicting 17 HP. Duncan's return stroke as it hits him glances off of the plant's tough integument.
Will resolve Duncan's next attack after the boards come back up.

Trample (Ex) As a full-round action, a creature with the trample ability can attempt to overrun any creature that is at least one size category smaller than itself. This works just like the overrun combat maneuver, but the trampling creature does not need to make a check, it merely has to move over opponents in its path. Targets of a trample take an amount of damage equal to the trampling creature's slam damage + 1-1/2 times its Str modifier. Targets of a trample can make an attack of opportunity, but at a –4 penalty. If targets forgo an attack of opportunity, they can attempt to avoid the trampling creature and receive a Reflex save to take half damage. The save DC against a creature's trample attack is 10 + 1/2 creature's HD + creature's Str modifier (the exact DC is given in the creature's descriptive text). A trampling creature can only deal trampling damage to each target once per round, no matter how many times its movement takes it over a target creature.
The last time, you hit rather more decisively, and the ascomoid was already wounded.

Also, I had edited my post from just over an hour ago to add the resolution of Duncan's second attack, but the boards went down before I could put it up. Here we go.
A pair of projectiles whizz past the ascomoid as it rolls over Duncan. Then, from underneath the bulk of the creature, the dwarf's axe flashes out and carves off a chunk of fungoid flesh.

Selveshanea was able to resist the ascomoid poison and pounces on the creature, but its claws are unable to penetrate. Ava charges into the fight and tears loose another piece of the ascomoid with her lethally-sharp teeth.
