Can anyone share with me how the rules for diseases work? Like what happens if they succeed the initial save what happens? What happens if they fail the initial save what happens? It says at Stage 2 the target is also slowed is that for one round?

All aflictions work like that:
When you are initially afflicted, you roll the save.
If you fail, after the "onset" period is over, you go to Stage 1 and suffer those effects.
If there's no onset, you suffer them immediately.
After the stage duration is over (noted in parentheses after the effects of the Stage) you roll your save again.
If you fail, you advance to next Stage (or repeat the effects of the stage you are if it's the last stage).
If you succeed, you reduce your Stage by 1 and take the effects of the new Stage (if you reduce to Stage 0 you're cured).
Critical saves/fails reduce/increase the amount off Stages you move by 1 more (so drop 2 stages or advance 2 stages)
Afflictions with the virulent trait need 2 successes to reduce a stage, or a critical save.

So, for your example:
After you fail your initial saving throw, since there's no onset period, you are Clumsy, Enfeebled or Stupified.
Since duration is 1 round, at the end of your round, you save again.
If you pass, disease is over.
If you fail you are also Slowed.
Second stage duration is again 1 round, so next round you save again.
If you pass, you're back to Stage 1 (so you basically lose the Slow but keep the other condition)(and repeat from there)
If you fail, you're no longer Slowed, but you're still Clumsy, Enfeebled, or Stupified, can't recover from those with normal means, and the next Save to reduce the affliction is the next day.
