I'm going to go back through Rotr with Agna(Ranger Deck), Ezren(Wizard Deck), and Flenta (Fighter Deck).
I haven't played a ranger in any of the APs yet and Agna appealed to me the most.She starts off being able to recharge animal allies and her high strength makes her better with longbows which I personaly favor over crossbows.
I chose Ezren because I wanted a dedicated caster as I haven't played one since my first run through Rotr when I played Seoni. Ezren is very different as he can't auto recharge his spells but he gets a higher spell count and I was intrigued by his transmogrifier role which Has some interesting item powers.
Finally I wanted to play Flenta because she seems like a lot of fun. She can auto recharge her weapons and with one of her roles she can auto recharge or aquire an item with the magic trait. I also figured she could inherit any spells Ezren collected but didn't need or want.

I'm really tempted to solo run a 6 man group of characters focused on their released path, in a path they weren't meant for, to up the difficulty.. So things from Shackles, Ranzak, Jirelle, and Damiel (Damiel wasn't really focused on the set, but Runelords doesn't really allow for potion-based combat). For my 3 Wrath characters, I'll have to see which 3 (Kyra and Seelah I'm sure) are heavily focused on demons and corruption.
If making half your powers worthless doesn't up the difficulty, I don't know what does!

Keep in mind that demons + undead isn't worthless in RotR, so Kyra might not be optimal. Plus, her second power becomes AMAZING in a world where no blessings are Corrupted.
So... I'd go:
Wardstone Sentry Seelah
Abyssal Walker Harsk
Corruptor Seoni
That should do it.
Lirianne would also work from S&S because RotR lacks firearms.

Yea I realized after the edit time was up that I somehow forgot Lirianne. Ranzak's replacement for sure.
I don't know the Wrath characters well enough to know who to pick without looking at them, so I just picked two that seemed to make the most (well, technically least) sense to run in a nearly demonless path.

Not sure how helpful my input will be since when I've done this I run four man parties through, but here's my input. I ran a four man 'Arcane' group through of Seltyiel, Melindra, Zarlova and Flenta. Seltyiel got mad at Flenta when she 'cast' her first spell and he called her out for a fraud, but that's neither here nor there. Group ran well Melindra and Zarlova might be two of my favorite characters in the game this far. Melndra runs as a cross between a rogue and a wizard. Strong Dex helps with lots of the RotR checks, just careful on weapons since not weapon prof till later (if at all). Zarlova recharge is op at times it feels as I hardly failed recharge checks. Added her for the arcane ability later to add spells and hardly needed it at all. Charisma/Diplomacy checks were my only real struggle with the group.
I decided to run RotR again with characters the wife and I didn't use for Wrath. Got through the B scenarios with a group of Adowyn, Crowe, Sharda and Amaryllis. So far Sharda is my struggle since the last two healers I played were SnS Lini and Zarlova who both can recharge attack spells on top of their decks.

Thanks for you guys thoughts. Right now I think I'm gonna do 4 characters. I like the idea of Flenta and Zarlova.
Funny how there's always the temptation to add one more character. I was going to take this group of Kyra (class deck version), Feiya and Arabundi through RotR, but at the last minute decided there was a Dex-and-Cha-shaped hole and dropped in Valendron.
More recently, I've just finished playing 6 characters solo (I had to try it once): Damiel, Tontelizi, Tarlin, Siwar, Amaryllis, Harsk (CD). Damiel and Siwar proved to be strong characters, in spite of the limited potions and almost total lack of Task and Skirmish barriers for them to get their teeth into. If anything it was Tarlin who was the weak link.

I would say to play through Runelords using whatever characters you think look interesting. It's a forgiving set that should help you get a feel for how fun a particular character is, and which characters could work well together. Tarlin, for example, might not have done so hot in that group above, but I've found him to be a key member of the group I'm using to go through Wrath solo (with Enora + Adowyn).
