Yes, such an item can be made. Without getting into the debate on how to calculate crafting costs based on which order you craft things in, here's a breakdown of the cost to buy such an item from a shop/vendor:
Belt of Strength +6 = 36,000 gp.
Belt of Con + 4 = 16,000 gp x 1.5 = 24,000 gp
Belt of Dex + 2 = 4,000 gp x 1.5 = 6,000 gp
Total = 66,000 gp.
The 1.5 fold multipliers are there because they are "additional abilities" added onto the most expensive item.

And if so how does the cost of that work building up from just one +2 stat item.
That's a custom magic item and as such is subject to GM discretion.
You would probably use the Multiple Different Abilities clause under Magic Item Creation to determine the price. Yes, enhancement bonus to different ability scores seems to be fairly similar, but that's how the Belt of Physical Might is priced in relation to the Belt of Giant Strength.
Belt of giant strength +6: 36,000 gp
Belt of incredible dexterity +2: 4,000 gp + 50% = 6,000 gp
Belt of mighty constitution +4: 16,000 gp + 50% = 24,000 gp
So you're looking at 66,000 gp assuming you don't increase the 3rd different ability at an extra 50%.
