If you're threatened by multiple characters does it add to your casting defensive concentration DC more than just 15 + double spell level? Is there a method where you combine factors to make one concentration check if you're under different conditions at the same time...like threatened + suffering continuous damage? Or, is it going to take multiple concentration rolls?

No, Casting Defensively is not affected in any way by multiple people threatening you.
Of course, if you do not cast defensively and prefer to take the Attacks of Opportunity instead, multiple people threatening you is bad news indeed.
Multiple conditions like the one you cited would require multiple rolls. Example: threatened and suffering continuous damage from Acid Arrow would require one for Casting Defensively and one for avoiding the last burst of continuous damage.

I'll just take this opportunity to spread knowledge of an interesting tactic I've tried for in-melee spellcasting:
As a move action, pull a rock out of your pocket. This will provoke.
As a free action, drop said rock. It's done its job.
As a standard action, cast your spell. No need to cast defensively because everyone's used their AoO for the round. ;)
I've done this twice so far, and haven't regretted it. Do NOT do it all the time, but there can be certain circumstances where it's a good idea.

I'm also not clear why this tactic is better than "take a 5-foot step". Also, do this once, and a smart attacker knows to not use his AoO on you pulling out a pebble.
You might be cornered, or you're already standing where you need to be in order to affect the desired area (and a 5ft step out of the threatened space would ruin that).
And yes, it generally works no more than once per combat.
Definitely do not make this your standard operating procedure. It's limited in reusability and very situational. It's just another trick in your arsenal, just in case you NEED to cast while threatened and that spell MUST go off.

You're correct that it's hilarious, but it goes more like this:
Me - "I retrieve an item from my person..." *use best "resigned to pain" voice* "...which provokes, so go ahead and take your AoO."
GM - *resolves attack*
Me - "Now I drop the worthless pebble I pulled out, and cast BLAH".
GM - "What was the rock for...?"
Me - "To soak the AoO before casting."
GM - ".........That's cheating!" Alternately, they just sit in stunned, narrow-eyed, open-mouthed silence for a few seconds.

I provoked AoOs on purpose to be capable of casting in the same turn. I also provoked AoOs from enemies on purpose to allow adjacent allied caster to cast before the beginning of the enemy turn. GM wasn't especially surprised by this. Nor was I when someone tried the tactic of soaking AoOs for the casties.
