I would say no, there is no penalty.
The context you didn`t mention to your question is the ´-4 penalty for humanoids who don´t have 2 hands free´ issue, presumably. The thing is that the Grapple rules don´t specify ´free´ FOR WHAT. Brass Knuckles let you carry items in that hand (and if you aren´t carrying anyt hing, your hand would be free in that sense), but you can´t WIELD a weapon in that hand. Nothing in the Grapple rules suggests that the ´free hand´ is in reference to being able to WIELD an item, so I would defer to saying that Brass Knuckles don´t interfere with Grappling. And after all, it would be VERY common to have 2 Brass Knuckles, one on each hand, which would prevent Grappling COMPLETELY if one assumes that being unable to WIELD a weapon counts as not having a free hand for purposes of Grappling.

I think that's a bit of a stretch. If you don't have enough control to wield a weapon you don't have enough control to get a grip on someone/something.
