How can I make sure I get an e-mail notification when someone replies to my thread or post on the forums?

You can’t. Because the forums and store are so integrated, they decided that getting auto-relegated to spam folders for sending out too many emails to an isp in a small timeframe would not be good, so never spent the time to develop this. The Paizo forums are like 90% custom coded in-house to support this integration, so they need to prioritize their development time, usually meaning that the forums get whatever time is left over from the store and download sections of the page
You won't get a notification, but you can use the Focus option to make it quicker for you to check if there have been any responses when you visit the forums. If you click on "FOCUS" at the top right of the forum screen near the search box, the display will only show you threads that you've read or posted in, arranged so that the most recent activity is at the top of the screen.
