Does the enemy just need 1 rank in Spellcraft to be able to identify Ill Oman is being cast on them? What would the DC be? And could I be able to make a knowledge check on my enemy before I used Ill Oman on them so I wouldn't need to waste it if they did have ranks in Spellcraft.

1 rank is enough to attempt to identify the spell, though they might fail--the DC is 15 + spell level. There's no practical way to tell whether the enemy has ranks in Spellcraft (well, there's probably an obscure spell somewhere), but if they're a spellcaster odds are good that they do. Even non-casters occasionally put a point in to have a shot at identifying subtle effects.
