For example, if an archer character enters a well lit room that is 100ft long, 25ft wide. The last 25ft of the room is completely filled with 24 zombies, one each square surrounding a Ghast.

Questions
1) Can the archer shoot the Ghast even if 4 zombies may be in front of him. Will the Ghast just receive a +4 to his AC

2) Could the Ghast decide to crouch / hide to prevent being hit by arrows.

3)Could the Ghast aim a spell at the archer, or would the 4 zombies in front block the spell attempt.

4) If the Ghast was behind Large creatures like an Ogre, would this make a difference

1) Yes
2)GM could call that improved cover and grant a +8 to AC, but it's not total cover. GM could also interpret it as still just soft cover and only grant +4 to AC.
3) Yes, but if it's a ranged attack spell the zombies would grant a +4 AC bonus to the archer.
4)Not really.
