That will depend on your GM and his campaign world. It may be a requirement for entry into a guild of assassins, or it could be part of a ritualistic experience the character needs to hone his skills. There are many different ways to run that requirement and that makes it highly unique to each game.

Hi.
I'm new in this world, just started last week. So, reading the Core Rules find the Assassin Class very interesting but says that I need to kill somebody to become an Assassin as a prerequisite.
Who I need to kill? An NPC, a Player's Character, a friend, a dog...?

First and foremost you should make sure your GM is okay with allowing Evil aligned characters. That is definitely not the norm for most tables due to the issues that crop up.

Whoever they say to kill.
Assassins, typically, are daggers-in-the-back for hire. You don't get to choose who you kill.
As a genral test, expect to be asked to kill a friend (PC character, important pc, etc.) or someone completely innocent (little old lady, kid, pretty girl, etc.). The general point of this prerequisite is to prove that you are able and willing to kill, and typically they want to make sure that you will not get any 'morals' getting in the way. They want to make sure you are evil.
So don't expect to be given a first job where the taget is the kind of guy that just asks to get assassinated (corrupt official, son of someone powerful flaunting his power to hurt people, general murderers, etc.). No, those are easy- good-y two shoes would be able to do it if he psyched himself out. No- they want to make sure you can keep to the job, even when it is disgusting.

For the Assassin PrC, you need to kill someone for no other purpoe than to become an assassin. Essentially, it's a cold blooded murder of anyone you want or are told to kill.
What you want is probably the Slayer class. They have no Evil only restriction, full BaB, Sneak Attack, an ability that gives them increased damage and skill bonuses versus their specified target(s), and probably most relevant to your interests, at 10th level can take the Assassinate Advanced Talent.
Assassinate is, simply put, a much, much, MUCH better version of the Assassin's Death Attack (it only takes one round of study instead of 3...which is a lifetime, and does the exact same thing otherwise).
Same flavor if you want, a more Neutral or Good oriented flavor if you want that instead, and a very easy to build class that doesn't involve any fiddly multiclassing, especially with PrCs that have weird Special prerequisites and skill ranks and so on (good for your first character, in other words).

+1 for go for some option other than the painfully horrible Assassin Prestige Class.
I always wonder how many wannabe assassins kill someone for free and then are told, "just kidding, chump, ha!-ha" in a Nelson-from-the-Simpsons sort of way by the Assassin's Guild (or a NE Practical Joker's Guild).
