Most likely, it is a build that also has a lot of levels in master spy (which also gets death attack, and its levels stack for the DC).
With that, you can get up to 15 stackable levels, for a total of 25+INT DC. So maybe
1-5 (whatever- maybe something martial like a ranger?)
6-7 Assassin
8-15 Master Spy
16-20(whatever combo of assassin and master spy levels).
Will it be effective to play? Probably not, since the death attack is delayed during the master spy levels. But it is the highest DC.

A Slayer is a good base class, as it gets sneak attack and things that synergize with Assassin. The Assassination Advanced Slayer Talent is better than Death Attack, though. It only takes one turn, and Studied Target adds to the DC. Most of the Assassin's features can be replicated with Slayer Talents, except Uncanny Dodge, Hide in Plain Site, and a faster sneak attack advancement. A Slayer with a couple of levels of Shadowdancer probably works better than Assassin overall.

Hmm. White-Haired Witch, into Feral Gnasher, Suffocating Strangulation feat.
One grapple check, then immediate Constitution checks, or the enemy falls unconscious and goes to 0 hit points.
Once you get Anaconda's Coils, retrain Witch level.
Add Skill Focus(Stealth) and Hellcat Stealth, to hide without magic, in an open field, on a bright sunny day.
