Does Spell focus add a +1 to the the trip attempt of a magic missile enhanced by Toppling metamagic?

No, spell focus only affects DCs of spells. Improved Trip would give you +2 on trip attempts, though.

Thanks! Then lastly would spell specialization give you +2 since it counts the spell as if you were 2 levels higher, and you trip based on CL+Casting Stat

Should, yeah. Anything that explicitly increases your caster level for the casting of this spell would modify it in all appropriate ways as though you where a higher caster level. All caster level determined variables would be effected.

If the check is CL+stat, and you increased your CL with a feat, by leveling, with a magic item etc, then... you would have a higher caster level for that roll too.
