So let's say the target fails his saving throw and drops his item. Next turn, he makes the save to pick it up. Does he have to make another save to try to use it? Or is the "saving throw made in each round" for both of those things?

I would say that he makes one saving throw per round ("each round").

One per round. If he has dropped it and then picks it up, he could then use it that round without a second save.
A fun tactic my kid came up with when she was first starting to play was greasing a weapon, then mage handing it away. Only works with non-magical items weighing less than five pounds, but a fun low level tactic nonetheless.
