I have a question concerning the Tangleburn Bag (not Tangleroot) from Ultimate Equipment (pg 109):
Tangleburn Bag (150g)
This sack contains tanglefoot bag materials and alchemical
powders that burn at a high temperature. It functions like a
tanglefoot bag, plus a direct hit on a creature deals 1d6 points
of fire damage, and the creature must make a DC 20 Reflex
save or catch on fire. If it catches on fire, for the next 2 rounds
extinguishing the flames is a DC 25 Reflex save instead of a DC
15 save, and using water to extinguish the flames creates a
burst of burning material equivalent to alchemist’s fire making
a direct hit on the target (including splash damage). After the
initial 2 rounds, the flames may be extinguished as normal.
Crafting this item is a DC 30 Craft (alchemy) check.
- The spell "Create Water" can be tossed at Short range, so can it trigger the effect of the "alchemist's fire"?
- Would the "alchemist's fire" deal the extra 1d6 afterburn since it is considered a direct hit?
- Is there any reflex saves in relation to the "alchemist's fire" damage?

1) Yes, it should.
2) Yes, the effect is equivalent to a direct hit and that should include afterburn.
3) No save to avoid initial damage, but the Ref save to extinguish the flames should apply as normal for alchemist's fire (though I think the DC will be 25 instead of 15 as long as you're within the 2-round intense burn window of the Tangleburn bag.)
So using Create Water after throwing a Tangleburn bag (or tossing a bucket at it) does indeed give you a bit more punch for your item, at the expense of an additional action.

Question: Is a Holy Water Flask enough to extinguish the fire and / or trigger the extra effect? I could understand if it spread the flames because there is water present. But is it enough to douse the flames? Just wondering.

I would think not, but ask your GM. I don't think there is an 'official' volume of holy water per flask.
