Drugs are probably beyond the target age range for the game, so are probably out. If they exist they will need significant benefits and drawbacks to make them believable.
Poisons I thing would be pretty easy to implement, and if they don't come at launch they will almost certainly come when implementation of assassination as a mechanically supported feature is added.

There is something really wrong with the concept that drugs are out due to "target age bracket" while poisons are ok. The same thing as sex is bad, but massive violence is ok.
However, I hope they have mechanics for food, drink, alcohol, pesh (drugs), and many different types of poisons. These things exists in the Pathfinder RPG and there are even gods that have these things as profiles. The game also has an alignment system that should allow for a more mature audience.

From Targeted ESRB Rating?:
Until and unless we decide that we need to make a retail product, and get pushback that such product must be ESRB rated (or PEGI rated in Europe), and we decide that it's worth the hassle to comply, we'll probably sidestep it.
ESRB rating or not, Paizo will not be approving content that would be inappropriate for the average 13-year-old.

From that same thread:
What you can assume is that the visual depictions of violence and sexuality, among other things, will be no more explicit that the illustrations you currently find in Paizo's own Pathfinder books, which we believe are acceptable to market to an audience ages 13 and up.
If it's in the books, it's allowed in the game.

Well, according to the Wikipedia (I know, I know) the "Use of Drugs" descriptor can be found on Teen rated games. Poison I don't think is too much of a concern - WoW bears a teen rating and features a fully functional poisoning system for the rogue class, which at one point did include the creation of such as well.
