Yes. But what would be the point? Usually the rune is placed on some writing that is meant to remain secret. Destroying the paper ruins all the other writing on the paper. Disabling it allows everyone to read the paper safely. Also when the detonation occurs the paper is destroyed (unless it can somehow resist the 6d6 fire damage).
Burning the paper would effectively destroy the spell since it can only trigger on being read. But it destroys the paper too, making it very hard to read.
