At 3rd level, a horizon walker learns total dominance over one terrain he has already selected for terrain mastery. When dealing with creatures native to that terrain, the horizon walker treats his favored terrain bonus for that terrain as a favored enemy bonus (as the ranger class feature) against those creatures. This bonus overlaps (does not stack with) bonuses gained when fighting a favored enemy.

Great. Got it. But what if you’re a Guide? Let's say you have Terrain Dominance in 'Desert'. The Guide 6/Horizon Walker 3 is fighting a creature native to the Desert. If we assume his Favoured Terrain bonus is at +6, can he then designate the creature as his ‘Focus’ and receive an additional +4 bonus to attack and damage rolls for a grand total of +10 against this creature?

I would say yes to the +10, the focus is not a favoured enemy bonus, its a misc bonus which will stack.

RAW yes, RAI...maybe.

While we're on Terrain Dominance. As I see it: "We're in a desert. This creature lives here. TD applies." is one way to bring the ability into play. Another is "We're on a mountain. This creature is a Foo Beast. According to the bestiary, Foo Beasts live on mountains. And in deserts. TD applies." Is another way, and also valid.
