This is a very good question, though I would say being that it is a Optional Rule that the answer would be no.

My understanding matches Dragnmoon's that the optional massive damage rule is not used in PFSOP.

I ran 6 tables at a con this weekend and I would say that about half the fights would have been over in one round if I followed this rule for NPCs.

"optional" rules, as printed in the books are generally not approved for PFS. Not optional in the sense that all the rules are optional, but those that are specifically listed as optional in their description.
With the propensity for PC/NPC power-attacking raging barbarians, smiting paladins, empowered fireball tossing sorcerers, and such, it would probably create a lot more one-shots.
But probably a good question to clarify in the FAQ.

Really? They were doing over 50 points of damage in one hit consistantly the first round? Wow.
Massive Damage (Optional Rule): If you ever sustain a single attack that deals an amount of damage equal to half your total hit points (minimum 50 points of damage) or more and it doesn't kill you outright, you must make a DC 15 Fortitude save. If this saving throw fails, you die regardless of your current hit points. If you take half your total hit points or more in damage from multiple attacks, no one of which dealt more than half your total hit points (minimum 50), the massive damage rule does not apply.
We used this in our last home campaign, and it actually killed some things maybe half a dozen times in the 15 levels the characters played through (And I had some really hard hitting players, let me tell you...). Usually if something has enough hp's to survive a 50+ hit, then they'll only fail the DC 15 fort save if you botch the roll. YMMV
