First off, does a construct count an an object, or as a creature for the purposes of discern location? Does it having an int score change which it is considered?
If an intelligent construct has been touched, (or someone gets a piece of it), can it be protected by a mind blank spell?
Same question above, but for an intelligent item, such as an item familiar.
Is there any way to protect a non-intelligent construct/item which has been touched from being located?

I believe that a construct is a creature.

Mindblank is an abjuration and I see no reason why a construct, intelligent or otherwise, would not be affected by the spell. Mindless constructs would gain no real benefit from it, but they could still be targeted by it.

The first thing that comes to mind is you could put it inside a Mage's Private Sanctum area.

Discern location was changed for PFRPG, as was mind blank, I suggest checking them out:

Evidently, constructs can be both:
A construct is an animated object or artificially constructed creature.
I would let that line stand for its literal self. Any construct that is simply some item thta is animated through a spell like Animate Object would be treated like an object. Any construct that is built from the ground up to be creature-like would be an artificially constructed creature and would be counted as a creature.
So you could locate a golem after having only seen it, but you would have to touch an Animated Shield first before you could discern its location.
I see nothing in the rules for constructs or for Mind Blank that says you cannot cast a Mind Blank on a construct. Mind blank does say the target must be a creature, so that means animated objects are out, but artificially created creatures should be valid targets.
The level 3 Nondetection spell may be what you're looking for here to prevent more ordinary means of detection. It's not complete protection, but few things are.
As for protection from the specific 8th level Discern Location spell,
Nothing short of a mind blank spell or the direct intervention of a deity keeps you from learning the exact location of a single individual or object.
So no, since you can't mindblank an object, you better get your deity on the batphone and call him up quick to intervene, otherwise any old archmage with this spell who has touched your object will be able to discern its location.

Maybe not.
This is the classic Irresistable Force vs. the Immovable Object.
Mage's Private Sanctum says no form of scyring works. Discern Location says nothing short of Mindblank or a deity can prevent it from discerning an object.
Who wins?
I see two solutions:
1. let the higher level spell win. This would be Discern Location. With this ruling, 1 casting of Discern Location would see right through the Mage's Private Sanctum and find the object.
2. let them neutralize each other. The Irresistable Force clashes with the Immovable Object and both cease to exist. Now the item is no longer protected by the Mage's Private Sanctum, but the Discern Location failed to find it. If the caster whips up another Discern Location, or any other scrying, before the item's owner protects it with a new Mage's Private Sanctum, then the second divination spell will find the item.
I can see arguing it either way, but I'm fond of the first way myself.

Your Option 1 is how I always ruled it.
And since mind blank specifically foils discern location, I mischevously introduced a unique artifact into my campiagn world that can discern location with the additional "rider" effect of penetrate mind blank -- meaning that no one is fully safe from spying, as long as that artifact remains in existence.
