I wish to know one thing, if a caster casts underwater, with the DC= 20+ level of the spell and failed, he lost his spell or not?

To the best of my recollection, anytime a caster fails a concentration check, the spell is lost.

That's exactly what I also believe.

Is it a Fire descriptor spell? If so it fails to function. If it's any old spell there'd be no Spellcraft check unless you're in rough or stormy water which I would say is the same as vigorous or violent motion. If you can't breath water then you can't speak underwater clearly enough to cast spells in "strong voice."
