Try The Lost Goblin in Raleigh. There was a sign up on their boards looking for some people to do Pathfinder games.
A friend of mine knows the owner and talks with him on a regular basis. Personally I have only been there once, I live in Fayetteville, and I thought the store was a little sparse. The owner, Rich, is a nice guy from the 20 or so minutes I spent there. From what I gather a lot of his sales come from people ordering specific items. He also said he has the largest NC preorder of 4th edition of any gaming store. Only the big book stores have him beat.

Lost Goblin link is probably your best bet, but you might also try SciFiGenre in Durham link. They have a very active gaming area.
Staff at both places are extremely helpful and friendly.
Unfortunately, Lost Goblin Games has closed. I went by the other day and there was a salon in its place. Sad times.
