Yes, yes, and yes.

Here are some examples:

The Mobhad Leigh in northern Varisia directly connects the surface to Sekamina.

The Earthnavel in the Realm of the Mammoth Lords directly connects the surface to Orv (to the vault of Deep Tolguth in particular).

There's a route from Nar-Voth under western Osirion that leads down throuh Nemret Noktoria down into Orv (to the Black Desert).

Also : The Ghost Path is a semi-legendary pathway that leads from Minkai down into the depths of the Darklands. The Ghost Path is a series of ledges and caves that lead from an extinguished caldera on Mount Kumijinja in the Ikkaku Peninsula into Sekamina. The Ghost Path ends in an immense shaft that is the only place in the whole of Sekamina that is exposed to direct sunlight.

Isn't there also a huge waterspout that leads up from Orv as well? I can't remember its nam but I think it's located in the Sightless Sea.

Indeed. That is the Braid. Goes up directly from the Sightless Sea through the Inverted Sea to the bottom of the Arcadian Ocean.
