I don't plan to make it mandatory for scenario success, but I'm kicking around an NPC villain who, knowing the kind of devastation a combined arms party of Pathfinders is capable of doing, will challenge the PCs to a duel over the MacGuffin they're after rather than straight-up fighting them for it. I just wanted to know if using the optional mechanics on a per-scenario basis was a done thing. Thanks. :)

Yes, but. :)
One of the complaints about season 5 was the inclusion of secondary systems. It sometimes seemed like every second scenario had an influence track or a ticking clock or a obscure optional rule system.
So you may see some resistance.
Not to say you shouldn't do it, it sounds cool, but you should be warned.

Remember, for the open call it's a 2000 word quest. There's not going to be a ticking clock or anything; one of the guys at the end will be a gentlemanly sort who will issue an open challenge to the PCs to fight a duel with one of them, positing it will be better all around this way than to get everyone killed in a bloody fracas. Of course, they're perfectly free to not accept, and the concept doesn't necessarily require the UC duel mechanics either; I just thought it could be a nice touch if I could fit it in the word count.
