Havent seen an official answer to this.
However I would say it does what it says it does.
"to defeat a henchman or villain"
To me that would mean the combat check against any card with the tag in the upper right 'Henchman' or Villain'
I dont have a skelington Horde Barrier in front of me, but as for closing a location? no that is a check to close a location, you could use it to defeat the summoned henchman if that is what the location closing tells you to do, but not in a roll against the location itself.
So TLDR:
Yes, if a henchman is summoned i would say it is still a henchman.
