Provided the people are within the proper distance and are aware of each other, do barriers affect telepathy?

I would say no. Divinations don't need line of sight to detect something at the other side of a barrier and spells that can be blocked mention explicitly what kind of barrier would block it.
Also, the most similar spell would be sending, which also completely ignores any kind of barriers.

So I would say nothing blocks telepathy. (Except spells or supernatural effects that specifically do so.)

Nope, there's been no such restriction placed on telepathy, unless magical barriers are used.
