I am running my second game of RotRL today and the last session ended with the players hearing about Ameiko's disappearance.
Now, page 27 (Anniversary Edition) mentions an argument between Ameiko and Tsuto. Is it mentioned anywhere what this was about? I can't find it.
And another (maybe related?) question, is Tsuto's father mentioned anywhere? I have an Half-Elf in my group, so I had the idea of connecting Tsuto and that character, that might create an interesting situation in the Glassworks, half-brother vs. half-brother or something...

I don't believe the AP mentions who is Tsuto's father. Obviously he is an Elf. That should give you freedom to take it whatever way you want. Some lusty Elf for years has been roaming the countryside impregnating Human women? It is up to you. (I am glad you have the spoiler's warning.) I have read where in some campaigns they made Justice Ironbriar Tsuto's father. I thought that was a clever twist. But your PC may not want a villain that your party may one day have to kill as their father. That is unless you want some kind of Darth Vader angle.
Concerning the argument between Ameiko and Tsuto, the one at the Turandarok Academy, we can only guess. But it would likely have to do with Tsuto's anger toward Lonjiku, may be even murderous aspirations. Or it may have had something to do with Nualia.
Let us know what direction you take.

OK, the family's finally awake, so here we go (hoping that Paizo doesn't mind me quoting Paizo books on Paizo messageboards):
"When she was 13, Ameiko tried to reconcile the bad blood between her brother and her father, but the attempt backfired. Tsuto struck her in a fit of rage, and Ameiko as so betrayed and distraught by this that she ran away from home to Magnimar for several months. When she learned of her mother's death in a fall from the cliffs near her house, though, Ameiko returned home to Sandpoint. She found life at home more unpleasant than ever, but when another family argument broke out at her mother's funeral and Tsuto left town, Ameiko resigned herself to staying in Sandpoint to care for her father.
Ameiko left home for the second time at age 16 after life in the house alone with her father and the help grew too depressing to bear..."

Second on NobodysHome. The fact that Tsuto wasn't Lonjiku's son caused a lot of tension in the household, and Lonjiku had Tsuto sent to the local orphanage in Sandpoint. Ameiko would sneak over there to hang out with him without her father knowing, and they were actually fairly close.
After the reconciliation attempt, they didn't really see eye to eye anymore, and shortly after their mother's death Tsuto left town anyway (after his argument at the funeral with Lonjiku which ended with him beating Tsuto with his cane).
Now I'm going to spoilerize this next part, because its actually big backstory reveal that I read in the original Runelords, but sounds like may not be right there in the AE.
Now as for the second part, I don't think its mentioned officially anywhere who she cheated with. I do know (and possibly it has something in Jade Regent #1) that Lonjiku was traveling a lot early in their marriage, and Atsuii was lonely. Also, Lonjiku perhaps was sleeping with Chelaxian noblewomen. We won't get into morality or anything, but she eventually sought solace somewhere. She never reveals who it was. Heck, she could have slept with the Mierani diplomat who lives in Korvosa. Perishial I think. Otherwise, I guess you could make it up.
