The Inner Sea Guide is the best place to start. It does not cover all of Golarion, but covers a good portion of it. It does not cover Tian Xia the Golarion analogue of East Asia (China, Japan, etc). However, it does cover most of what you need to know to play in Golarion.
You can also check out the Player's Guides to each of the Adventure Paths. These are free and are intended to give the players resources needed to play in the different areas of Golarion that are covered by each Adventure Path. As a side effect, they give an idea of the flavor of that particular part of Golarion.
There is NOT a single book that covers everything at this time. This is due to the fact that Golarion is still being written/ created. I belive that James Jacobs has stated that he has a beach ball in his office that he is drawing the Golarion globe on over time.
The Inner Sea Guide gives you an overview of all of Golarion that was written at the time it was published, so is the best single resource for getting to know Golarion.
Another place to look to get a good feel for Golarion is the Web Fiction. This is also free, and free is our friend. The fiction can give you a taste of what it is like to live in different parts of Golarion.

We're deliberately not attempting to detail the entire planet, because by focusing more precisely on one region (as presented in the Inner Sea World Guide), we can give the level of detail one needs to have for a campaign setting. We can't do that as well if we're trying to cover an entire planet.
Inner Sea World Guide is absolutely the place to start getting into Golarion if you're a GM or want to get into the setting.

The one thing I would like to see, which may already be available somewhere, is a map showing how the different parts of the world connect. The Inner Sea region is well covered, but how does it connect to the Tian side of the world? What about other areas? One big, not too detailed global map would really help with the big picture.

Ahh, cool. I actually just bought that book the other day, but haven't had a chance to go through it yet.
