The spellcaster roles the check to overcome his/her enemies SR. At the minimum, the bonus includes the spellcaster's caster level. Things like Spell Pentration and Greater Spell Pentration add to this number, as do items that increase your caster level (like the orange iounstone) and class features (like the archmage's Spell Power +1).

I've always had doubts about how the SR checks work. Is the spellcaster who rolls a d20 to beat the opponents' SR or the opponent who rolls to resist?.. and what are the bonus for a SR check?..Plz guys, I need the answer here, cuz I couldn't figure it out how it works in the books.
The simple way to think about it is as Spell Resistance = Spell AC.
The attacking spellcaster rolls 1d20 + Caster Level + Feat/class bonuses (as already mentioned above) vs. the "AC" of the Creatures SR (Spell Resistance). Then, like in combat, if you meet or excede the SR, your spells works on that creature.

Summary: If a monster has spell resistance it'll be listed in his statblock. To affect such a monster with a spell you must first roll 1d20 plus your caster level, and try to get it higher than the value of his spell resistance. If you succeed, the spell works as normal; if not, the spell fails. Of course, if the spell allows a saving throw, the monster still gets that.
