I am to for the first time Game Master Rise of the Runelords for two of my friends. I am to having Game Mastered before, but only brief modules and homebrewings. I have yet to reading through the whole thing and prepare maps, but our first session is on Friday so I have time. What is the best advices for running this for the first time?
What should I doing to accommodating two players and prepare? What is the biggest thing I should to be prepared for?

With only two players, you need to look very carefully at action economy, and particularly at effects that take out a single player on one saving throw: a failed save vs. sleep could result in a TPK very easily. (Eryllium and Nuala are the two encounters you most have to worry about here.) You might consider letting your players do two characters each, or having someone along to help share the burden.
At least read through the first section (Burnt Offerings) and have a clear idea of the plot so you know how things fit together, what the major NPCs are doing, and how they might react if things go pear-shaped. (Which they will, as soon as PCs get involved.)

I would also add in some NPCs to help out. Stat up a guard or acolyte to go with them, Ameiko after she's found in the Glassworks could help out. Use it as a story hook for her desire to capture Tsuto (if escaped) or right whatever wrong he caused to the town after finding out about the raids. And Shalelu would be a great help in Thistletop. You could tweak her down a level so that she doesn't take all the kills, but with the bigger baddies, she could definitely help out in those fights.
