So what would happen if all the pcs were female? Would Shayliss even approach any of them? And if she did and convince one of the pc to fight the "rats", what would happen if Ven catches them. Would it warrant a lower bluff check. Or would you simply change Shayliss gender but how would this change Ven reaction to the pc fighting "rats"?

Depending on your group, you could play the situation several ways.
Shayless might be sexually interested in a female PC, with her father's disapproval caused by the homosexuality or because the target of Shayless' affection is a "shiftless adventurer".
Some players wouldn't respond to that. For them, Shayless might adopt a form of hero worship, hoping to eventually get away from Sandpoint and "see the world". Instead of going to the basement for romance, she wants to show off a secret stash of gear, items saved for the day when she finally gets out of Sandpoint. Her father would disapprove of anyone "encouraging" his daughter to run off.
In my games, Shayless was eager to get out of Sandpoint, eventually running off to Magnimar, where she became a recurring character. One group's drow sorcerer had an ongoing relationship with her, while the paladin in the other group tactfully disentangled himself from the affair.

I asked basicaly the same question last year when we started Burnt offering and James Jacobs sugested that Shayliss should be open to alternatives... Her dad, not so much!
She ended up going for the dwarf in my campain. One of the other (female) PCs would have worked but she was already involved in something else (nudge! nudge! wink! wink!) and I didn't want to pile it all in on one player.

If your group is comfortable with it, then absolutely Shayliss would approach a female PC. As a general rule, we don't nail down any NPC's sexuality, unless that aspect of their character is a legitimate part of that character's story and personality. And even then, just as you can alter an NPC's race or class or gender to fit your world and play style, feel free to alter their sexual preferences as well.
Certainly, Papa Ven can be just as scandalized and angry no matter WHO he catches his innocent, beloved daughter in the cellar with!

We actually changed our game where Katrine Vinder was seeing Harkers neice Jessie and Vin doesn't like Banny because he permits it. Shayless' reputation is uber well deserved in this game as she, to quote OOTS, is "True Neutral".She has at the end of Burnt Offerings offered to every PC (2male/1female/and 1 "true neutral" male) none have been so foolish as to take her up on it.
