Mystic Theurge.
Although Cleric 3/Wizard 17 or Cleric 17/Wizard 3 are pretty close.
A cleric 3/wizard 3/Mystic Theurge 14 would be fun if it gave 9th level spells at character level 20.
Maybe that bard/druid Focloccan Whatchamacallit. Lyrist?
Maybe a Dragonslayer PrC? Or other monster slayers? Basically, you need to kill a whole bunch of stuff to qualify for a class that makes you really good at killing a whole bunch of stuff.
Essentially, I think there should be PrCs that actually require you to accomplish something and truly be prestigious to qualify for.
Maybe a Master Thief that needs to steal X gps, or succeed on Y number of schemes, capers, or grifts. Or a magician that creates a number of new magic spells or magic items. Or a tomb raider that explores a bunch of dungeons, or a mountain climber that has climbed all 7 celestial peaks, or a diver that has swum in all 11 sacred lagoons, or a master linguist that learns a dozen languages, or a planar voyager that visits all 4 elemental planes, etc. etc.
Maybe something like a Giant Weapon Wielder, where you struggled through several levels wielding a weapon that's literally too big for you to be good at, suffering Disadvantage on the majority of your attack rolls, and, once you qualify for the PrC, you finally get the knack for swinging giant weapons around, and even learn giant weapon tricks, like hitting 2 creatures with one swing, or hitting the ground and making shockwaves that knock over your opponents.
Maybe an advanced soldier that combines the Battlemaster fighter with the College of Valor bard. Basically getting higher level combat maneuvers, and maybe more uses for Bardic Inspiration, and definitely combining them in some way.
Maybe a Two-Weapon fighting specialty class that eventually learns to make 2 or more attacks as a bonus action. And learn how to wield Versatile weapons with one hand but dealing two-handed weapon damage. Maybe additional OAs.
It's kind of hard to come up with concepts that aren't better modelled using Feats and/or Multiclassing and/or developing new Archetypes.

I did up a feat that basically lets you be a Mystic Theurge, which I would show you, but it is unfortunately part of a product that I am working on. Suffice to say, I think that it could be handled rather well as a feat.
SmiloDan, how would a Cleric 3/Wizard 17 work? You'd only get access to 2nd level Cleric spells that way. Or is that what you meant?
I like the idea of a prestige class being the route for very focused characters, and even the way that people can customize after character creation without necessarily multi-classing into other entire classes.
