#1 Hydrate, Hydrate, Hydrate. Drink a ton of water. Tasty beverages with salt in them, while being fun will dehydrate you fast at Gen Con. So, for every 20 ounce Dew/Pepsi/Coke you have, follow it up with 60 ounces of water.

#2 Pack protein. Your going to be pushing yourself hard all day, have something with protein in it to keep giving your body the right kind of fuel to use. Candy is great, but the crash behind it is hard.

#3 HAND SANITIZER. People are dirty dirty dirty. Always sanitize your hands starting a slot, ending a slot, after leaving the bathroom. I burn through the stuff at big cons. I learned my lesson after contracting whooping cough at a Gen Con.

#4 Get a lot of sleep. I've been the snoring room-mate, I've been with a snoring room-mate. Ask your hotel staff for ear plugs, sleep masks etc. I've been there when someone wouldn't turn out the lights, or snored. Wear a mask and some ear plugs. Just have something loud enough to wake you up in the morning.

#5 As above. Bathe, and please please please - wear comfortable clothing, no one has come to see the dark side of the moon. So please make sure your clothes do their job ^_^

Most of all, have a good time. Because this is fun for everyone. Enjoy yourself, make new friends, see old friends - and go out and Murder, Death, Kill some bad guys!
So, since this thread has been hijacked, allow me to send out something I used to send to my judges back in the day.
