Would it be legal to take all the cards for the locations used, mix them all up, then deal 9 cards to each location, then mix in the Villain and Henchmen cards?
This would mean a location may not have the correct number of certain types of card listed.

Example: The Academy has 4 spell cards. By mixing the cards the way I said above, there is a chance the location could only have 1 or 2 spell cards, or less. The missing spell cards would be in other locations, including places that may not normally have spell cards.

Why would you want to do that? One of the best things about knowing how many cards are in each location is so you can plan your assault. Start off with a location with few barriers/monsters so you can build up your decks for the tougher locations.

Well, clearly it wouldn't be "legal" per the rules, but I don't see how it would break the game if you did it that way as a houserule. I agree with TClifford though, I think it's more fun if you have some idea of what's available in a given location.

Randomizing things that way removes some of the flavor from the game as well. The Temple, for example, is supposed to be a (mostly) safe place where you're likely to gain a blessing—a good place to regroup if you've lost a bunch of cards—and the Nettlemaze is supposed to be a dangerous monster-filled spot—a place where you might have to prep before you visit.

There are also some mechanical elements that might get messed up a bit; for example, if there are no monsters in the Warrens, it'll be much easier to deal with than it oughta be. (And there are more locations in future chapters where their powers care about their deck contents.)

I think it would be neat to do on a custom scenario, but I agree with the others and wouldn't try it if I'm going through an adventure. Might be fun to do one day if you're getting bored of the scenarios and want to randomize the game a lot.

a random location huh, like instead of a card list you take say 3 of each type of card that can go into locations, shuffle them and then deal out 15 random cards to the location instead of having assigned card types... now that could be interesting. the location could be some weird chaotic vortex place so you never know what will be in there from game to game...

so many good ideas that can work with this game, I see a long play-life for me, I hope it is doing very well that they might consider a more robust release schedule next "season" instead of just a base set and one adventure path.

You may enjoy Chapter 3.

hahaha I am already enjoying the BLEEP out of this and it's just the base stuff, so I know you guys have only got more imagination and idea's to show us!
