I want to give the players of my next game an opportunity to make friends with one of the combatants I have set up. I already have plans for what will happen if he joins the party.
If they manage to defeat the main bad guy that is forcing this guy to attack them then (before they defeat this guy) he will stop attacking them. If they knock him out first then there is a small but possible chance they will heal him awake (as the main baddie won't be there to question). If either of these happen then negotiations should occur.
He will start of indifferent or unfriendly if they knock him out. Either way his inclination right then is to leave. What would it take mechanically to get this character to join their party?

I see diplomacy as the key. Charm person or intimidate can get them to help temporarily. My question is more what numbers do I need set the checks at. I want to give them penalties and bonuses based on how they RP.
What level?
If you want it to be mostly based on their roleplaying but still want rolls in there, I suggest starting at DC 10 or 15 and then give bonuses or penalties based on what they say. Make it several checks to make it interesting.
I would do away with the rolls though and just let them TALK to each other.

Legendary Games put out a product that seems exactly what you are wanting.
I am assuming you wanted a mechanic more expanded than faking a diplomacy roll.

I agree with Powerdemon do away with the roles and let them talk it out.
if you want them to be inclined towards friends make him fight softball
attack the high AC guy, don't Coup de Grace ignore the squishy.
if they think about it will seem like he was pulling his punches and it will help to endear them to him.

If you really want to do this, then allow the player to make a check of some after certain events. I wouldn't use a skill, since that could be seen as manipulation of sorts. If I decided to do this, it would be a Charisma check DC 15 (or higher if appropriate). Allow a check after a period of positive roleplay combined when they spend together. Feel free to grant a +2 bonus for special circumstances, such as if the character has risked harm to help to NPC, has made a sacrifice, has kept an important secret, they have common life experiences, hooked up NPC with Seahawks tickets at 50 yd line, etc. Don't allow it more than once a week. The DC is hard enough that it won't seem like a random success, but easy enough that a persistent player will be able to pull it off. Also, once the first success is made, there could still be additional checks to go further.
Generally though, just keeping it to role-play would be ideal.
