Can a character prof with a thrown one handed weapon make it large and use it as a Lg Thrown Weapon (-2 attacks for being Large on med character) throwing it 2 handed with 1 1/2 times str added?

Well, you aren't allowed to buy a large version. How are you planning to make it large?

In general, I would say no. A large weapon would be too unwieldy to throw effectively without further specialized feats.

Large versions aren't always available, but they are, in fact, available for purchase.

Thrown weapons, regardless of their handedness, only add the Strength modifier once and require a full-round action to throw. With the feat Two-Handed Thrower, however, your damage jumps to 1.5 Str and you can throw as a standard. You'll take the -2 penalty to attack rolls from the size difference, but nothing prohibits you from throwing large weapons.

Whether or not this is a viable tactic I leave to you to find out.
