Enlarge Person gives a definite +2 Str regardless of what size you are ending up at. That would not stack with Monstrous Physique II's +4. The end result of these two would be +4 Strength -4 Dex, -2 Hit, +2 CMB, +2 CMD, -2 AC, +4 Nat AC and of course Huge size which comes with reach benefits. So it's not bad but might not be worth the 2 Standard actions to cast them both.
