I was thinking of ways to utilise this, and considered the option of putting it into a spell-storing ioun stone, then having cast it from the ioun stone when, say, I want my Overland Flight spell extended. Not necessarily the best use of the spell slot, but this is as an example.
I realise that the casting time would increase to a standard action if the spell is cast from an ioun stone. Would this stop the spell from affecting my other spell if my familiar readies an action to activate the ioun stone?

I can't see why it wouldn't work. If your familiar uses the stone you aren't casting it on yourself, which looks to be the only restriction. If you are using a readied action the standard action casting time is irrelevant.
But..
I could see this being ruled against you as well, especially if your GM thought it was 'cheesy'. There's not really a precedent in the rules either way because immediate actions are never compared to readied actions so we have no idea how they interact.
