Can a character having 6 xp play a Tier 1-2 scenario before leveling up to 3rd level, or must the character level up once having 6 xp before it is played again?

No, you automatically level up as soon as you gain the XP needed to level up. You can't choose not to level them up.

You were level 2 when you had 5 xp. The moment you earned your 6th xp you became 3rd level.
